Gene expression patterns of two DNA replication related proteins at various growth stages and UV damaging conditions in a planktonic diatom / 浮游矽藻中核酸複製指標蛋白在各族群生長階段與紫外線損傷下的基因表現

DNA polymerase α and proliferating cell nuclear antigen (PCNA) are two DNA replication-related genes. The main purpose of this is to distinguish which gene is suitable for the application in estimation of phytoplankton population growth rate. A diatom (Skeletonema costatum) is used in this study. The mRNA content of these 2 genes was detected by competitive quantitative reverse transcription-polymerase chain reaction (CQ-RT-PCR) at various growth stages and different UV-treatment conditions. During exponential phase, population growth rate reached 0.6 day-1. The level of DNA polymerase α mRNA ranging from 600 to 1200 molecules cell-1 detected in cells from exponential phase, is higher than the level of 0 to 500 molecules cell-1 found in cells from the stationary phase. Similarly, PCNA mRNA level was between 900 and 3500 molecules cell-1 during exponential phase but decreased to 0 to 500 molecules cell-1 in stationary phase. In UV treatment experiments, DNA of S. costatum was severely damaged by UVC treatment and a large quantity of thymidine-dimers was formed. The mRNA concentration of DNA polymerase α and PCNA were high in the UVC-treated samples. The results indicate that both DNA polymerase α and PCNA were involved in repair of DNA damage. In UVA/B treatment, T-dimer formation was less obvious mRNA concentration of DNA polymerase α and PCNA were similar in both UVA/B-treated and control samples. In contrast, the level of PCNA protein seemed to increase in UVA/B-treated samples. Since UVA/B is the major form of UV radiation in the natural environment, our results suggest that both mRNA of DNA polymerase α and PCNA are suitable indicators for estimation of population growth rate of S. costatum.
