Inhibition of histamine production in mackerel flesh by Lactococcus lactis subsp. lactis CCRC 14016 / 乳酸乳酸球菌CCRC14016抑制鯖魚肉組織胺形成之研究

The Investigation was conducted to figure out the causes that Lactococcus lactis subsp. lactis CCRC 14016 can inhibit the formation of histamine in mackerel fillets. Firstly, series of storage test of treated fillets were carried out. Mackerel fillets were treated by soaking in L. lactis subsp. lactis CCRC 14016 cell suspension containing 3﹪salt for 24 hr, and then by blow-drying for 8 hr and finally by storing for 20 days. All the treatment and storage were proceeded at 4℃. The pH and histamine content of the fillets soaked in cell suspension were significantly lower than those in cell-free supernatant (p<0.05) after 20-days storage, and the histamine content of the former treatment was not increased; but the volatile base nitrogen (VBN) had no significant difference between the two treatments. L. lactis subsp. lactis CCRC 14016 could not degrade histamine, but could metabolize histidine into ammonia. By using enzyme-substrate test, we found nisin could not inhibit the activity of histidine decarboxylase. After the termination of the storage, some histamine-producing bacteria were isolated from mackerel fillets which were soaked in cell-free supernatant; on the contrary, no histamine-producing bacteria were found in the fillets soaking with L. lactis subsp. lactis CCRC 14016 cell suspension. Furthermore, L. lactis subsp. lactis CCRC 14016 could reveal inhibitory ability against histamine-producing isolates, including Enterococcus cloacae, Lc. delbrueckii delbrueckii, Pediococcus acidilactici, Lc. lactis subsp. lactis and Morganella morganii CCRC 11257. The growth of these histamine-producing organisms was not inhibited but enhanced by 1:10 diluted L. lactis subsp. lactis CCRC 14016 cell-free supernatant. We suggested that suppression of histamine produced in mackerel fillets could be causes of reducing free histidine that is a substrate to form histamine and of inhibiting some histamine-producing bacteria by L. lactis subsp. lactis CCRC 14016.
