The aim in this study is to produce the N-acetylchitooligosaccharide mixture with immune-enhancing ability by cellulase digestion of shrimp chitosan, followed by acetylation. Twelve samples of chitooligosaccharide mixtures, obtained from shrimp chitosan (with 90 % deacetylation) digested by cellulase at concentration of 6、8 or 10 mg/mL for 1.5、2、2.5 and 3 h, were evaluated for their immuno-function on cells of human hybridoma HB4C5, human-like macrophage UMf, and mouse-like macrophage J774.1. The chitooligosaccharide mixture from cellulase digestion at 8 mg/mL for 2.5 h showed the highest enhancing activity on the proliferation of HB4C5 and UMf cells, which were 1.7 and 1.3 times, repectively, compared to the control. Sugars with 1-8 degrees of plymer (DP) were found in this chitooligosaccharide mixture and the pecentage of sugars with DP≧6 was 22.6 %, which was the highest among the twelve samples tested. This mixture was further acetylated with acetic anhydride, in oder to obtain samples with acetylation degrees of 10、50、75 and 100 %. After reevaluation of these 4 oligosaccharide mixture samples, the sample with 100 % acetylation (A 100) demonstrated the greatest positive effect on cell proliferation. The levels of serum IgG and IgM were similar for mice injected intraperitoneally with this chitooligasaccharide mixture (A 100) and N-acetylchitohexaose compared to those without injection. In addition, this chitooligosaccharide mixture (A 100) enhanced the Con A and LPS-induced proliferation of mice lymphocytes. By Con A induction, the proliferation rate of lymphocytes by 1.6 and 1.4 time, respectively, for A 100 and N-acetylchitohexaose injected mice By LPS induction, the proliferation rates were similar for A 100 and N-acetylchitohexaose injected mice, both which were higher than control.
