1.Effect of high glucose on inositol and taurine transport
It has been known that the intracellular concentration of inositol is low in type II diabetes mellitus(DM)patients. This may be caused by patient’s hyperglycemia status or cellular mutation of inositol transport .The relationship of inositol and taurine transport in normal or type II DM patients’red blood cells (RBC) was investigated in the thesis. We find that high concentration of glucose inhibit inositol transport in normal RBC , but this inhibition is not such obvious in type II DM patient . In low K+ concentration , the degree of glucose inhibition on inositol transport is significantly higher than in normal K+ concentration . Regarding to taurine , we find that high glucose may promote taurine transport in a concentration-dependent manner. However, we did not find similar results in Type II DM patients’ RBC . Taken together , inosital and taurine may via different transport pathway in normal and Type II DM patients’ RBC.
2.Inositol and taurine transport in human leukemia cells
Many researchers have suggest that inositol and taurine may transport through cells by the same pathway under hypotonicity, and this pathway may be a Cl- channel . We used tamoxifen (Tx)、5-Nitro-2-(3-phenylpropylamino) benzoic acid (NP)、and PD89059(PD), three chemicals which have been demonstrated to be effective anion channel blockers, to investigate insotiol and taurine transport in human leukemia cell . The results show that inositol and taurine transport systems are inhibited by these three chemicals in a similar degree. Thus , we suggest that inositol and taurine may via the same volume-sensitive pathway under hypotonicity in human leukemia cells.
