Using mt DNA to Study the Phylogeny of Doratosepion (Cephalopoda:Sepiidae) and Evolution of Sepia Genus / 應用粒線體DNA探討Doratosepion(Cephalopoda:Sepiidae)之分類地位及烏賊屬之演化情形

碩士 / 國立彰化師範大學 / 生物學系 / 89 / Since the nineteen century, the classification of Doratosepion has been changed several times. In the twenty century, Doratosepion was classified as a genus, then it was classified as a subgenus, a subfamily, or a group under subgenus. In 1998, Khromov and others, based on the cuttlebone and the external features, classified it as Sepia (Doratosepion), one of the six species complex of Sepia.
In 1990s, due to the development of molecular biology technique, many researchers used the 16S ribosomal DNA (16S rDNA) and cytochrome c oxidaseⅠ(COⅠ) to solve the phylogenetic problem of cephalopods. In this study, regions of two mitochondrial gene (16S rDNA and COⅠ) are used to compare with the morphological data in order to clarify the taxonomic level of Doratosepion. The molecular data are also used to investigate the evolution of Sepia.
The samples are collected from Taiwan, Australia, and France. By the analysis of morphological character, the morphological phylogenetic tree is constructed; by the analysis of molecular character, the molecular phylogenetic and evolutionary trees are also constructed. Judging the results, both 16S rDNA and COⅠphylogenetic trees support Doratosepion as a monophyletic group and should be a species complex belongs to Sepia, and Sepia is very likely the earliest taxon of Sepia genus.
