碩士 / 國立海洋大學 / 水產養殖學系 / 89 / This study investigated the protective effects of experimental vibrio vaccine on the grouper (Epinephelus sp.). The agglutinating and bactericidal activities of grouper antisera against Vibrio alginolyticus, and its inhibitory ability against protease of the extracellular products (ECP) of the bacterium were also examined and analyzed. The V. alginolyticus S4y strain, originally isolated from moribund grouper were passed in the fish to increase its virulence re-identified, and used in this study for experimental vaccine preparation and lethal challenge.
In single immunization tests, only treatments using lipopolysaccharide (LPS) and/or those with adjuvant exhibited good protection. The ECP preparation did not show protective effect as expected. The mortality of all the treatments increased at week 8 indicating that the protective duration seemed to be short.
In oral booster tests, different vaccine preparations and delivery routes all exhibited enhancement of fish immunity against lethal challenge S4y strain. In addition, after oral booster for four weeks, the fish were protected until week 12, the end of the experiment.
The inhibitory abilities of fish antisera from all the treatments against extracellular protease of S4y were lower than those from control. This inhibitory ability of fish antisera from all the treatments was decreased as duration increased at different vaccines treatments. This ability may be simply due to non-specific immunity since the agglutinating ability was apparently decreased at week 12 after oral booster for 4 weeks.
The bactericidal activities of fish antisera, obtaining from fish at week 8 or week 12,added with fresh complement against S4y strain were detected. However, this bactericidal activities were not detected in fish antisera obtaining from fish at week 4.The reason may be due to lower titer of antibody present in fish after vaccination of only 4 weeks
