Detection of Aeromonas hydrophila in food by an anzyme-linked immnuosorbent assay / 以酵素結合免疫分析法檢測食品中水生產氣單胞菌

碩士 / 國立海洋大學 / 食品科學系 / 89 / Aeromonas hydrophila is widely distributed in various foods and can cause gastroenteritis in human. The aims of this research are to establish an enzyme-linked immunosorbent assay (ELISA) and apply this ELISA to detect this microorganism in milk, oyster, and ground pork. The female New Zealand rabbit was immunized with the extract of surface antigens of A. hydrophila 8-169, which was isolated from oyster, for the production of antibody. An ELISA with sensitivity of 105 CFU/mL was estabilshed by using the antibody and enzyme-antibody conjugate at 10 mg/mL. Food components from milk, oyster, and ground pork caused the decreases of ELISA absorbance. The high contaminants in the produce of oyster and ground pork in market caused the increase of ELISA value. The interference from food components could be decreased by washing and heat treatment. However, heat treatment also resulted in the denaturation of the surface antigen of A. hydrophila. Accordingly, ElISA value decreased after heat treatment. The interference from food components could be effectively decreased with 5-10 times of dilution. A. hydrophila spiked in commercial pasteurized milk can grow well over 107 CFU/mL at both 28oC and 4oC. Similar results were obtained for A. hydrophila in oyster and ground pork at 28oC. However, the amount of A. hydrophila did not change at 4oC because both of oyster and ground pork were highly contaminated by this microorganism. The ELISA established could detect both the naturally-contaminated and artificially-spiked A. hydrophila in milk, oyster and ground pork. The results from ELISA and the plate colony count were similar. A little higher value was obtained from ELISA due to the presence of no-heat-caused dead cell in food such so in ground pork.
