Micrococcus sp. strain SDC07 was isolated from the biofouling material a glass slide that was suspended in sea water, and it performed aggregating mucous colony morphology on the marine agar. Eight saccharides were used to replace the carbon source of MSBB (mucus screening basal broth), the results indicated that while strain SDC07 growing in MSBB with the original sucrose preformed higher EAS () yield (0.54 ± 0.23 g/L). As to the effect of sucrose concentration (1.0-9.0%) on the EAS yield, the data showed that as the sucrose concentration increased to 3.0% the EAS production by the strain SDC07 was the best (1.47 g/L). Among eight combinations of nitrogen source were evaluated, the highest EAS yield was observed from 0.5% polypeptone (PP) + yeast extract (YE) group which is the N-source of the original MSBB, had 0.80 ± 0.41 g/L. Effect of PP and YE concentration on the EAS yield, the result presented the replacement of 0.46 PP + 0.06 YE was higher 0.96 ± 0.05 g/L EAS yield. The addition of 2% disodium-β-glycerophosphate in MSBB achieved a highter EAS yields (2.18 g/L) by the strain SDC07. Among the addition to four metal ions tested, Mn2+ gave the best yield (2.25 g/L). The effect of cultural condition was examined individual, the results showed that while the initial pH at 5.2, shaking speed with 150 rpm, or incubation temperature at 26oC, the highest EAS yield range 0.88-1.16 g/L.
To test the effect of different substratum on the EAS powder yield from strain SDC07, substratum such as activated carbon (AC) flake, chitin flake, wood piece, stainless steel beads, glass beads, or polypropylene beads were added to modified-MSBB-M for the examination. The chitin flake added to modified MSBB-M performed the highest EAS yield as 1.03 ± 0.27 g/L than other testing substrate, the second was AC flake which obtain 0.98 ± 0.14 g EAS/L broth). The compositions of EAS lyophilized powders that obtained from culturing strain SDC07 in the MSBB and modified MSBB-M, the former moisture content 11.45%, by dry weight, total sugar content 77.17%, curde protein 11.20%, ash 7.79%, curde fat 0.80%, the latter moisture content 4.4%, by dry weight, total sugar content 83.89%, crude protein 8.65%, ash 4.60%, crude fat 1.64%.
The results on the solubility of rehydrated aqueous solution with various concentration of EAS used was dissolved distilled water, 1-5% NaCl, 80% formic acid, and 99% dimethyl sulphoxide. And it was partial dissolved in 95% isopropanol, and 90% ethyl ether, although it was not soluble in 95% ethyl alcohol, 95% propylalcohol, and 99.5% acetone. The RV (relative viscosity) of EAS rehydrated aqueous solution was increased when the concentration of EAS was increased in the reacting solution. However, EAS solutions with the addition of increasing NaCl concentration were performed lower RV. The 0.2% lyophilized EAS powder rehydrated solution showed the higher and more stable RV over a pH range from 6.0 to 10.0. The RV of 0.2% lyophilized EAS powder rehydrated solution increased as temperature was stabled from 10 to 20oC, then the RV was decreased as the temp higher than 20oC. Effect of EAS addition on emulsifying activity and emulsion stability while mixed with isolated soy protein, the result showed that 0.2% EAS emulsifying activity was the best, emulsion stability of the EAS from strain SDC07 was lower than that xanthan gum had. The 1.0-3.0% strain SDC07 EAS rehydrated soltion had presented the capability of film formation.
