Biocontrol of Rhizoctonia solani on phytopathogenesis of mung bean using Cellulomonas flavigena NTOU 1 and shrimp-shell powder / 以產黃纖維單胞桿菌配合蝦殼粉防治立枯絲核菌對綠豆之病害

碩士 / 國立海洋大學 / 食品科學系 / 89 / This research was conducted to evaluate the practicability of Cellulomonas flavigena NTOU 1 to be the biofungicide to inhibit the growth of plant pathogen Rhizoctonia solani. In the test of complex media containing colloidal chitin, C. flavigena could significantly (p<0.05) inhibit the growth of R. solani, resulting hyphal hydrolyzation under the observation of SEM. In basal medium containing dried mycelia (0.02% w/v) as sole carbon source, C. flavigena could be activated to induce the production of N-acetylglucosaminidase (GlcNAcase) and small amount of N-diacetylchitobiase (chitobiase). Addition of shrimp-shell powder (SSP) in the media could enhance the activities of GlcNAcase and chitobioase. However, the addition of SSP in soil could reduce the mortality of C. flavigena. In mung bean pot experiment inside incubation chamber, addition of SSP (0.1% w/w) in natural soil without sterilization, the inoculation of C. flavigena (1.7×107 CFU/g soil) could significantly inhibit R. solani causing plant disease (p<0.05). Addition of SSP (0.1% w/w) in natural soil could enhance the proliferation of total viable bacteria. C. flavegena could also inhibit the growth of Penicillium spp. and Fusarium sp. isolated from soil. Besides R. solani, C. flavigena could also inhibit the growth of Colletotrichum gloesporioides, Alternaria brassicae and Fusarium oxysporium pathogenic fungi.
