Infectious pancreatic necrosis virus (IPNV) belongs to the Birnaviridae family of virus and has bisegmented-double strand RNA genomes, which is also one of the contagious and widespread fish diseases often causes severe damages to the aquaculture in Taiwan. The present study was attempted to investigate (1) whether IPNV-E1S could induce apoptosis in the zebrafish liver epithelium cell line (ZFL) at 18 and 28C during different culture interval (24 and 48h), and (2) the regulatory mechanism of apoptosis through caspase by the measuring caspase 3 and 8 activities.
IPNV-E1S could infect ZFL at 18C and induce the apoptosis of ZFL, the cell death rate was 50~65% and over 80%, after 24 and 48 h-culture, respectively. The viral protein expression and virus titer analysis indicated the virus protein expressed and processed in ZFL, and the infectious titer increased after infection, suggesting the virus can be copied in ZFL and released in the medium. In contrast, the ZFL survived after IPNV-E1S-infection at 28C and the expression of viral protein was incomplete, which resulted in the decrease of infectious titer. These results indicated that the IPNV-E1S is susceptible to infect the ZFL cells at 18C. Furthermore, the morphological changes in associated with DNA damage (TUNEL assay) and the elevations of caspase 3 and 8 activities after IPNV-E1S-infection suggest the apoptosis of ZFL may be resulted from the death signal transduction by activating the caspases.
