碩士 / 國立臺灣大學 / 海洋研究所 / 89 / This study focuses on the pH and buffering capacity problems involved in the determination of ammonia in seawater by the indophenol blue colo-rimetric method. The pH-shift puzzle was unraveled by acid-base titration. The major buffering capacity for the indophenol blue method is induced by the combination of the phenate system (pK = 10) and a previous — un-known Mg-citrate system. By adding extra alkali reagent to compensate for the pH difference in saline media, the molar absorptivity was found to be 21000 /M/cm at 640 nm. For sample with a variety of salinities, an empirical equation is suggested to correct the results.
