碩士 / 國立屏東科技大學 / 熱帶農業研究所 / 90 / This study examines the activation response of Taiwanese abalone (Haliotis diversicolor supertexta) and uses different concentrations of immunostimulants of zymosan, phorbol myristate acetate (PMA), laminarin, sodium alginate and trypsin to induce the hemocytes of phenoloxidase (PO) and superoxide anion (O2-) of the animal. The result showed no response when using trypsin to stimulate the PO and O2- in the hemocytes of abalone, but a significant dose-effect with other immunostimulants. Among the immunostimulants used sodium alginate and laminarin provided the best induction effect. Their induced prophenoloxidase (proPO) was inhibited by protein enzyme inhibitor of soybean trypsin inhibitor (STI), p-nitrophenyl-p-gunidinobenzoate (p-NPGB) and benzamidine. The above facts indicated proPO the hemocytes of abalone processed the proPO enzymes system, which existed in an inactive form.
Using three different concentrations of superoxide dismutase (SOD) to clear O2- of hemocytes in Taiwanese abalone, it is found that there had higher clearance rate upon using 150 U/μ(51.69±6.58%) and 300 U/μl (60.08±10.25%)。
The second part of this study examines Taiwanese abalone exposing to 20, 25, 30 and 35 ppt salinity levels at varying time intervals (0, 24, 72 and 120 hour) for measurements of their immune disease-resistant factors (hemocytes of PO, O2-, phagocytosis and clearance efficiency) and total hemocyte counts. The result indicated that the PO, O2-, phagocytosis and clearance efficiency was significantly decreased (P<0.05) when abalone were exposed to 20, 25 and 35 ppt in 24 hours. Total hemocytes counts of abalone were significantly increased (P<0.05) when exposed to 35 ppt. The challenge test showed the mortality rate of abalone were significantly increased (P<0.05) when exposed to 20, 25 and 35 ppt, and reached 100% cumulative mortality rate when exposed to 20 ppt. Among the tested salinity levels, the immune disease-resistant ability of abalone was the highest at 30 ppt.
