Effects of NaCl on detached leaves of Pluchea indica (L.) Less / 氯化鈉對鯽魚膽切離葉片之影響

碩士 / 國立高雄師範大學 / 生物科學研究所 / 91 / The research was to investigate the effects of NaCl in detached leaves of pluchea indica from Wushanting Mud Volcanic area. Based on appearance, specific growth rate and TTC reduction activity, the salinity optimal for growth ranged between 0-250 mM NaCl. As compared to those at 250 mM NaCl, the detached leaves at high concentration of NaCl showed a decreased appearance, specific growth rate and TTC reduction activity. An increase in free proline level in detached leaves of pluchea indica caused high concentration of NaCl. The accumulation of proline by NaCl is relation to protein degration, but could not explaine by Δ1-pyrroline-5-carboxylate redutase activity and proline dehydrogenase activity. Exogenous application of proline had no effect of growth in detached leaves of pluchea indica. Sorbitol decreased the content of protein and increased the level of proline in detached leaves of pluchea indica at the concentration iso-osmotic with NaCl. The ativities of peroxidase and guaiacol peroxidase were observed to be less under recovery. 26KD protein was derived from SDS-PAGE at higher than 500 mM NaCl in salt treatment and recovery.
