In vitro Assessment of Immunomodulating Factor from Extract of Trionyx sinensis / 甲魚萃取物之生體外免疫調節功能評估

碩士 / 國立海洋大學 / 食品科學系 / 91 / The human diet contains a great variety of natural carcinogens, as well as many natural anticarcinogens. The physiological effects of food factors are of particular interest because may be useful for human cancer prevention. Techniques for human cell culture will be useful both for basic studies of cell biology and for evaluation of physiologyically active substance in vitro. Shellfish are popular source of traditional Chinese medicine. In this study, Trionyx sinenesis was selected for immunological function assessment. Trionyx sinenesis extracted with hot-water (100℃, 30 min) or 40% ethanol then concentration and ammonium sulfate saturation precipitation. In the order to clarify performance at the cellular level, human-derived hybridoma and macrophage were cultured and treated in a serium-free system. Hot-water extracts of Trionyx sinenesis promoted cell proliferation, immunoglobulin production toward human hybridoma cell lines HB4C5, and promoted cell proliferation toward HL-60, K562 immuno-cell lines. After purified by using Phenyl-TOYOPEARL hydrophobic interaction chromatography, the proliferating factor was gained from W40. In the prescence of 5% serum, the promoting factor induced nitrite production remarkably, and showed synergistic effect with INF-g. The promoting factor can promote the phagocytose of PBMC, the cytotoxic activity of PBMC to K562, and the ability of production cytokine from helper T cells. The 40% ethanol extracts can inhibite the growth of cancer cells such as breast cancer cells (MCF-7), hepatoma cells (HuH-6KK, HepG2) and colon cancer (HT-29). After purified by using Phenyl-TOYOPEARL hydrophobic interaction chromatography, the inhibitory affect fraction gained from E40. By using the DNA fragmentation assay, cell cycle determination and Annex V-FITC/PI assay, showed the cancer cell death induced by EPT0, which is necrosis not apoptosis.
