Immobilized invertase by calcium alginate / 以褐藻膠鈣固定轉化酵素

碩士 / 國立雲林科技大學 / 工業化學與災害防治研究所碩士班 / 91 / Invertase is an important enzyme in the food industry. It can hydrolyze sucrose into fructose and glucose, so called invert sugar. This research investigated the immobilization technique of invertase. Moreover, the immobilized invertase was packed into a column to form a continuous enzyme reactor.
Invertase is a glycoprotein with about 50﹪of carbohydrate moiety which may be oxidized by sodium periodate to generate aldehyde groups for covalent binding with carriers. The amino group of invertase can also be used for immobilization.
This research used calcium alginate as enzyme support. It’s structure、manufacture、diffusion behavior、physical and chemical property etc, have been studied by many researchers. Calcium alginate at different conditions were prepared for enzyme immobilization. Various support’s mechanical strength、diameter and density were assessed. Screening for potential materials and methods, were conducted. Optimal conditions for enzyme stability activity and high yields have been studied.
This research choose optimal conditions as Alg:2%(w/v), calcium chloride concentration:4%(w/v), soak time:30 min. Its density is 1.03 g/cm3, ﹪deformation is 23.40%, and diameter averaged at 2.71mm. Optimum conditions for continuous operation are: temperature 56℃, pH=4.5, substrate concentration 1.0 M, flow rate 1 ml/min.
