p73 is one of the p53 family members which are transcription factors involved in the regulation of cell proliferation, apoptosis and differentiation. p73-deficient mice exhibited severe developmental phenotypes including neurological defects and chronic infectious problems. It was shown that p73 was required for survival and maintenance of neurons in both central and peripheral nervous system. To investigate the role of p73 in lower vertebrate development, we cloned the p73 cDNA from zebrafish previously. During embryonic development, p73 transcripts were detected from the zygote period to the early larva stage. Whole-mount in situ hybridization revealed that p73 was expressed in most parts of the brain, including olfactory bulbs, telencephalon and hypothalamus. Expression of p73 was also detected in the primordium of pharyngeal arches and the nose. Moreover, p73 protein was found in the ovary and testis by immunohistochemical staining. Knocking down of p73 by antisense morpholino oligo resulted in several morphological defects in embryos in a dose-dependent manner, including delayed epiboly, shortened and curved body axes and smaller brain. During convergence and extension stage, p73 morphants displayed less extension in AP direction and less convergence in dorsolateral direction in their dorsal fates. Besides, no difference in apoptotic cell number was detected during this stage by TUNEL assay. Notably, skeletal defects including loss or malformation of pharyngeal arches were observed in 5-day-old morphants. These data clearly demonstrate a role for p73 in specific developmental processes of zebrafish. Nevertheless, the detailed mechanisms await to be elucidated by further experiments.
