Gene cloning and protein expression of a rice trehalase / 水稻海藻糖酶之基因選殖與蛋白質表現

It is commonly considered a storage compound but more recently has been recognized to function mainly as a protectant for maintaining vital structures in the cytosol under stressful conditions such as extreme temperatures, drought, and desiccation .It might do so by stabilizing membranes and protecting enzymes. Trehalose is also expected to be used for various applications in the fields of food industry ,cosmetics and pharmaceuticals. Trehalase catalyzes the hydrolysis of trehalose into glucose. It plays important role in regulating trehalose content in vivo and can be used for the analysis of trehalose in vitro.
In this study , a trehalase gene (Os-TRE) protein was isolated from rice encoding a 563-amino-acid. The predicted molecular weight was 62 kDa and its pI value was 5.49. Os-TRE gene was overexpressed in Escherichia coli system and the recombinant protein was shown to possess trehalase activity. This is the first plant trehalase gene which was successfully expressed as an active recombinant protein. In the future, a antisense RNA technology or RNA method can be used to suppress trehalase activity, which might increase the trehalose content in rice plants and confer tolerance to different abiotic stresses .
