Cloning and expression pattern of FOXO5 in zebrafish / 斑馬魚之FOXO5基因的選殖及其表現模式

碩士 / 中山醫學大學 / 生化暨生物科技研究所 / 93 / The superfamily of Forkhead transcription factors consists of approximately 100 members and is expressed in an array of species ranging from yeast to human. Previous report has demonstrated that each members of FOXO family show a tissue-specific and developmentally specific expression pattern, which is involved in pattern formation of the embryo. FOXO family transcription factor can regulate a wide range of gene expression including cell cycle-related genes, DNA repair genes, and energy metabolism genes. When different FOXO members phosphorylated by PKB inhibits the activities of FOXO family transcription factors. Phosphorylation of the FOXO is required for cytoplasmic localization, possibly by binding to 14-3-3 proteins that render the proteins to be exported from nucleus and tether them in the cytoplasma。 14-3-3 proteins expressed abundantly in brain region, which can interact with many proteins to modulate a lot of cellular processes including cellular signal pathways、cell cycle progression and apoptosis. Binding of 14-3-3 can protect the target protein from degradation, alter the sub-cellular localization of target protein, and alter the intrinsic catalytic activity of the target protein。 In this study, we have investigated the expression FOXO5 in different stages and organs of zebrafish embryo and to determine whether 14-3-3 γ protein induced sub-cellular localization of FOXO5。We have performed RT-PCR to detect the expression pattern of FOXO5 in 0 to 96 hours post fertilization. Expression of FOXO5 was found in brain, olfactory lobe, lateral lines, and gill by using in situ hybridization. In addition, the cytoplasmic localization of FOXO5 was observed only in co-transfected with constitutively activated form of Akt/pEGFP-FOXO5 / HA - 14-3-3 γ into human lung cancer cell line H1355.
