Utilization of inorganic carbons by Chlorella FJ03 / 小球藻FJ03對無機碳資源化利用之研究

碩士 / 輔仁大學 / 生命科學系碩士班 / 93 / Recently, using organism to fix carbon dioxide has been respected gradually. Biological utilization of carbon dioxide is to make it as carbon source of organism for growth, for example the photosynthesis of plant and microalgae convert carbon dioxide to carbonhydrate. The purpose of this experiment is to isolate the microalgae from nature environment which can fix carbon dioxide and to estimate the possibility of carbon dioxide fixation in photoreactor system of the microalgae by medium design and growth factor effect combine with growth kinetic model and biophotoreactor design. The following are result of this experiment: the microalgae, Chlorella FJ03, isolate from environment can be used as a model for carbon dioxide fixation study. After studying of Chlorella FJ03 optimal culture there is a result of medium composition: NaNO3 0.5g/L, K2HPO4 0.06g/L, MgSO4•7H2O 2.0g/L, CaCl2•2H2O 0.06g/L, citric acid 0.06g/L, ferric ammonium citrate 0.25g/L, EDTA 0.001g/L, Na2CO3 0.02g/L and trace metal solution 1ml/L. In the test of different carbon dioxide concentration, Chlorella FJ03 have optimal growth condition in 5% and 10% carbon dioxide. In the test of light intensity, Chlorella FJ03 can grow better in high light intensity. In 200L biophotoreactor Chlorella FJ03 have higher growth rate than in simple experiment. Growing Chlorella FJ03 in 200L continuous culture model and controlling ratio of medium supplying in and flew out at 8.34L/h can make Chlorella FJ03 to maintain the cell concentration at 0.7 in OD600. Using 200L biophotoreactor to precede continuous carbon dioxide fixation and control the cell concentration at 0.7 in OD600, carbon dioxide concentration at 10%, air explore at 0.616L/sec. In these culture conditions, the efficiency of carbon dioxide fixation of Chlorella FJ03 is 0.371g/L everyday. Each day in this culture can increase the dry weight of cell in 44.235g.
