Effects of Nonylphenol on Spermatogenesis in Male Swordtail Fish ( Xiphophorus hellerii ) / 壬基苯酚對雄性劍尾魚(Xiphophorushellerii)生精作用之影響

We used methods of DRAQ5 for evaluating spermatogenesis kinetic and 7-AAD for analyzing germ cells apoptosis by flow cytometry. We evaluate various germ cell populations and apoptotic germ cell to know possibly adverse effects in male swordtail fish ( Xiphophorus hellerii ) treated with different doses ( 0, 90, 360 and 720 μg/L ) of 4-nonylphenol ( NP ) at 14 and 28 days various post-treatment time. Treatment of fish with high dose of NP have trend toward decrease gonadosomatic index ( GSI ) at day 28. The expose of NP resulted in significant changes in the relative percentage of different germ cell populations as a consequence of which the elongated spermatids/spermatozoa ( HC ) declined significantly ( P <0.01 ) and raised synthesizing DNA ( S-phase ) population ( P <0.05 ) following expose to 360 μg/L at 14 days. The significant decreased in the relative percentage of elongated spermatids/spermatozoa ( HC ) ( P <0.05 ) after post-treatment 90 μg/L at day 28. The significant decline in the relative percentage of round spermatids ( 1C ) and spermatogonia/somatic cells ( 2C ) but significant raise in the relative percentage of spermatids/ spermatozoa ( HC ) following expose to 720 μg/L at 14 and 28 days. Furthermore, flow cytometric analysis germ
cells apoptosis on the testis of 360 and 720 μg/L post-treatment fish indicated reproductive damage especially in early apoptotic cells have significantly raised but relative percentage of viable cells have significantly raised following expose to 90 μg/L at 14 days. Histopathological analysis found has dose-dependent and adverse effects on the testicular structure. These included reduced number of spermatogenetic cysts, rupture spermatozeugma in the efferent ducts and inhibit the process of spermatogenesis. These results suggests that exposure to NP exactly can result injury the processes of spermatogenesis and decreased fertility in male swordtail fish.
