Effects of an Insecticide, Trichlorfon,on the Physiological and Immunological Responses of the Giant Freshwater Prawn, Macrobrachium rosenbergii / 地特松對淡水長臂大蝦生理及免疫反應之影響

碩士 / 國立屏東科技大學 / 熱帶農業暨國際合作研究所 / 93 / Giant freshwater prawns, Macrobrachium rosenbergii, at intermolt stage of 15 – 20 g, were exposed to different concentrations of trichlorfon (0, 0.2, 0.4 mg l-1). The parameters analyzed included physiological acclimation responses, pathogenic changes and immune responses. Also analyzed were sublethal concentration; hemolymph, muscle and hepatopancrea glucose and lactate contents; effects on glycogen changes; acetylcholinesterase activity inhibition; the histopathological changes; osmotic and ionic regulation, and the base-acid balance; phenoloxidase activity; respiratory burst activity; superoxide dismutase activity; total haemocytes; susceptibility; phagocytic activity and clearance efficiency. The results show that the LC50 at 96 hr were 0.255 mg l-1; and histopathological changes including hyperplasia, fusion and necrosis on gill lamellae were observed. Under different concentrations of trichlorfon, the prawn hemolymph, muscle and hepatopancrea glucose and lactate contents increased significantly; muscle and hepatopancrea glycogen also decreased significantly; and relative acetylcholinesterase activity were significantly inhibited following the treatments of different concentrations of trichlorfon at 24h; decreased osmolality, Cl-, Na+, pCO2, HCO3- and TCO2 levels of the hemolymph and increased pH and pO2 levels of the hemolymph were observed. In the parameter of immune response, hemolymph phenoloxidase activity and respiratory burst activity increased significantly; and superoxide dimutase activity significantly decreased, resulting in an accumulation of superoxide. However, there was no significant difference in total haemocyte counts. Phagocytic activity and clearance efficiency also decreased significantly. After trichlorfon exposures, the prawn’s susceptibility to pathogen challenges increased significantly. It therefore, indicated that trichlorfon treatments affect not only physiological parameters of M. rosenbergii through neurotransmission but also immune responses in disease resistance.
