Administrations of hot-water extracts of Sargassum duplicatum and Petalonia binghamiae enhance the immune ability of white shrimp Litopenaeus vannamei and its resistance against Vibrio alginolyticus / 重緣葉馬尾藻與小海帶熱水萃取物強化白蝦免疫力及抵抗Vibrioalginolyticus感染之研究

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 93 / This study monitored the immune response of white shrimp Litopenaeus vannamei ( 10.42 ± 1.39 g ) and its resistance against Vibrio alginolyticus when the shrimp were injected with hot-water extract of Sargassum duplicatum or Petalonia binghamiae at a dose of 2、6、10 or 20 μg g-1. The immune parameters included total haemocyte count (THC), phenoloxidase activity, respiratory burst (release of superoxide anion), superoxide dismutase activity, phagocytic activity and clearance efficiency to V. alginolyticus.
L. vannamei injected with the hot-water extract of S. duplicatum at 2、6、10 and 20 μg g-1 showed increases in THC、phenoloxidase activity, respiratory burst and phagocytic activity and clearance efficiency to V. alginolyticus after 1 day. In another experiment, L. vannamei which had been injected with the hot-water extract of S. duplicatum, were challenged with V. alginolyticus at 1.4×106 colony-forming units (cfu) shrimp-1 and then placed in seawater of 34 ��. The survival of shrimp that received the hot-water extract of S. duplicatum was significantly higher than that of shrimp injected with saline and the control shrimp at the termination of the experiment (6 days after the challenge).
In the case that L. vannamei immersed with the hot-water extract of S. duplicatum at 100、300 and 500 mg l-1 showed increases in THC、phenoloxidase activity and respiratory burst after 1 day. In another experiment, L.vannamei which had been immersed with hot-water extract of S. duplicatum, were challenged with V. alginolyticus at 1×106 cfu shrimp-1 and then placed in seawater of 34 ��. The survival of shrimp that immersed in hot-water extract of S. duplicatum was significantly higher than that of control shrimp at the termination of the experiment (6 days after the challenge). It is concluded that hot-water extract of S. duplicatum increased the immune ability of L. vannamei and resistance from V. alginolyticus infection.
L. vannamei injected with hot-water extract of P. binghamiae showed increased THC、phenoloxidase activity and respiratory burst after 1 day. L. vannamei injected with hot-water extract of P. binghamiae showed increased phagocytic activity and clearance efficiency against V. alginolyticus after 1 days. In another experiment, L. vannamei which had been injected with hot-water extract of P. binghamiae were challenged with V. alginolyticus at 1.4×106 cfu shrimp-1, and then placed in seawater of 34 ��. The survival of shrimp that received hot-water extract of P. binghamiae was significantly higher than that of shrimp injected with saline and control shrimp after 6 days.
The present study indicated that both hot-water extracts of S. duplicatum and P. binghamiae can enhance the immune ability of L. vannamei and its resistance against V. alginolyticus infection. Further research is needed to evaluate the immunostimulatory effect of L. vannamei and its resistance against V. alginolyticus when shrimp feeding a diet containing either hot-water extract of S. duplicatum or P. binghamiae .
