Toxic effects of copper sulfate on the physiology and immunity of white shrimp Litopenaeus vannamei / 硫酸銅毒性對白蝦生理及免疫機能影響之研究

碩士 / 國立臺灣海洋大學 / 環境生物與漁業科學學系 / 93 / The objective of this study is aimed to understand the toxicity of copper sulfate and its effects on the physiological and immune responses to white shrimp(Litopenaeus vannamei ). The acute toxicity of copper sulfate was experimented, and 50 % lethal concentration(LC50)was further calculated.。Physiological parameters of hemolymph lactate, glucose and triglyceride, and the immune parameters of the activities of phenoloxidase, reactive oxygen intermediate and superoxide dismutase were monitored for 48 hr under acute stress conditions.
The 50 % lethal concentrations of copper sulfate to the white shrimp was calculated to be 100.6, 65.6, and 50.4 mg/L at 48, 72 and 96 hr, respectively. The hemolymph lactate markedly elevated in 1 hr after exposure to copper sulfate in the control and all concentrations examined, followed by notable declined from 3 hr and thereafter. Similar increasing trends were also observed in the hemolymph glucose and triglyceride, marked increases were detected in 1hr and a high level of the response was maintained for a longer period. Both hemolymph glucose and triglyceride decreased down to nearly basal level from 6h after the exposure. The observations suggested the energy metabolism, both aerobic and anaerobic, was enhanced for sustained supply of energy needed for physiological compensation to the stress introduced and also maintainance of physiological homeostasis. When white shrimp (Litopenaeus vannamei ) expose to toxic copper sulfate of varying doses, the activities of phenoloxidase, reactive oxygen intermediates represented by superoxidase, and superoxide dismutase all decreased as the presence of copper sulfate, and the activities reduced as the stress prolonged. The observations suggest that all the parameters related to shrimp innate immunity all reduced with increased copper sulfate concentrations. The correlations among the physiological and immune parameters were further analyzed, and significant correlation among them were evidently demonstrated (p < 0.01).
