Effects of Heat Preconditioning and Heatstroke on Cellular Damages of the Hippocampus in Rats: S-100 and GFAP- like Immunocytochemistry and Western Blotting Analysis / 大白鼠海馬回經熱前制約及熱中風處理的細胞損傷:S-100和GFAP免疫細胞化學以及西方墨漬法

碩士 / 國立陽明大學 / 解剖暨細胞生物學研究所 / 93 / Heatstroke-induced neuronal damage can result from the heat and lower perfusion of cerebral blood flow. We investigate the morphological changes of neurons and astroglia in the CA3 and CA4 sectors of hippocampus and dentate gyrus in heatstroke-induced rats by suing immunocytochemistry, electron microscopy and Western blotting. In addition, we also evaluate the effects of heat preconditioning and glycerol injection on the neurons of the hippocampus. Our results indicate that the duration of the heatstroke induction is 99.8±8.3 minutes. A striking characteristic is that there appear the symptom of hyperthermia and hypotension in the heatstroke-induced rats. Moreover, we find that the reactive pyramidal neurons of CA3 and CA4 sectors show intensely methylene blue staining in association with the hypertrophyied cytoplasmic processes of the astroglia. Furthermore, the granule cells of the dentate gyrus show high tolerance effects in response to the heatstroke. The heat precondition may cause to activate the different signaling pathways via astroglia to mitigate the reactive cell changes of neurons. On the other hand, administration of glycerol can increase the survival time after the onset of heatstroke. Furthermore, we also show that the reactive astroglia contain the dilation of perikaryon and the hypertrophied processes. Such morphological features may be involved in the homeostasis of the perivascular space of the astroglia and the blood brain barrier of the blood vessels. In conclusion, our data suggest that the astroglia may play an important role in the protection of neurons against the stress and insult from the environmental toxicity, especially in the synapses of pyramidal neurons with granule cells of hippocampus.
