Development of the Recirculating Culture System for Rotifer / 輪蟲循環水養殖系統之研發

碩士 / 國立中興大學 / 生物產業機電工程學系所 / 94 / A rotifer’s recirculation culture system which could be used to rear the rotifer Brachoinus sp. was designed and developed in this research. The system was constructed and combined together with the parts of the bio-filter tank, the culture tank, the algal supply tank, the algal storage tank, the temperature control system, and the auto cultivating solution inlet and outlet control equipment. The four varieties of feeds, the alga Isochrysis aff. galbana, the baker yeast, the freshwater Chlorella powder and the PUFA emulsion were first selected as rotifer’s food for the batch culture test. The test with the alga Isochrysis aff. galbana held the highest growth density. The rotifer inoculated density, cell density of the alga Isochrysis aff. galbana, the feeding method and the temperature of the cultivating conditions to influence the rotifer growth were also studied. The experimental results of the fundamental batch tests showed that the optimal rotifer inoculated density, cell density of the alga Isochrysis aff. galbana, the feeding method and the temperature were 50 ind./ml, 100×104 cells/ml, one-supply and 35℃, respectively. The batch raising cultivation of rotifer with 12 and 60 liters were also conducted. The method of non-aeration was the more appropriate way obtained in 12 liters test. The 60 liters test could get the maximum rotifer growth density 1216 ind./ml as the feeding shock reduced. Two semi-continuous cultivating tests using the newly developed recirculating-water culture system with the hydraulic retention time (HRT) two hours and the daily dilution ratio 16.67%(10/60) were performed. The first experiment with six days of batch raising cultivation and eighteen days of semi-continuous cultivation could produce the harvested solution 10 liters with the daily average rotifer harvesting density 827 ind./ml. It also demonstrated that the bio-filter held the nitrification function and the concentration of NH4+ was dropped from 1.6 mg/l to 0.005 mg/l with NO2- reduced from 5 mg/l to 0.5 mg/l. The second experiment used timer to control the cultivating solution inlet and outlet electromagnetic valves and resulted in the automatic semi-continuous culture of the rotifer. The test was first with three days of batch raising cultivation and followed with twenty-three days of semi-continuous cultivation. The experimental result showed that the system could steadily produce 10 liters of daily harvested solution as the daily average rotifer harvesting density of 1099 ind./ml. During the cultivating period, the rotifer growth density could be maintained between 915 and 1450 ind./ml with the average growth rate 0.182 day-1. The experiment also verified that the automatic semi-continuous recirculating-water culture system presently developed in this research obtained the capability of stable and fine cultivating functions to produce rotifers.
