碩士 / 國立彰化師範大學 / 生物學系 / 94 / Acetyltransferase and deacetyltransferase are highly regulated enzymes which participate in gene transcription. The normal equilibrium of these enzymes in biological systems paly important roles in growth proliferation and differentiation in normal cell. An imbalance in enzymatic activitues could ultimately lead to cancer cells proliferation. Recently, there has been a continued interest in evaluating the effects of several acetyltransferase inhibitors including polyamine-CoA conjugates, peptide CoA conjugates, anacardic acid, garcinol, curcumin, and Cu2+-OP complex. In this study, we investigated the effect of two Histone Acetyltransferase inhibitors; curcumin and Cu2+ -OP complex on Zebrafish embryo development. The results showed that: (1) the concentration of curcumin’s greater than 5.0 μM could reduce the hatching rate and survival percentage of Zebrafish embryo; also a bent in juvenile fish rear was observed; (2) An increase in OP’s concentration delayed the hatching of Zebrafish embryo; bending of the rear was also observed in hatched juvenile fishes; (3) concentration of OP greater than 1.2 μM reduced the hatching rate; (4) An increase in Cu2+ concentration also decreased the hatching rate and survival percentage of embryo; bending of the fish rear was also observed; (5) 2.0 μM Cu2+ remained the normal Zebrafish embryo development in 20 μM OP. Besides, we also cloned the promoter of Zebrafish apoptosis protein CED-6, and inserted into pEGFP-1 reporter vector. We showed that both curcumin and Cu2+-OP complex could induced the expression of CED-6 promoter drived green fluorescence protein.
