碩士 / 國立屏東科技大學 / 食品科學系 / 94 / The aim of this study was to evaluate the protective effects of soft-shelled turtle liver powder (TLP) supplementation on carbon tetrachloride (CCl4) induced liver injury. Liver damage was induced in male eight-week-old Sprague-Dawley (SD) rats by an intraperitioneal injection of CCl4 (40% CCl4 in olive oil) by a dose of 2 ml/kg body weight (BW). Sixty rats were divided into five groups, each group containing 12 animals. These groups included (1) olive oil injection as experimental blank, (2) CCl4 injection, (3) CCl4 injection and 4mg/ml silymarin (200mg/kg BW) supplementation, (4) CCl4 injection and 0.1 g/kg BW of TLP, and (5) CCl4 injection and 0.5 g/kg BW of TLP supplementation. The CCl4 was given twice a week for 8 weeks. Liver injury was determined by measuring serum levels of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT) at 1, 3, 6, and 8 weeks after treatment. The level of lipid peroxidation in the liver homogenate were also measured at 8 weeks, when rats were sacrificed for liver sampling.
The results showed that CCl4 treatment induced a significant increase in serum GPT and GOT levels as well as a high lipid peroxidation was observed in the liver tissue compared with those in the blank group. The rats supplemented with 0.1g/kg BW of TLP did not show improvement on the CCl4 -induced liver injury and lipid peroxidation, while the 0.5 g/kg BW supplementation of TLP was markedly effective in decreases of the serum levels of GPT and GOT and lipid peroxidation in comparison with those in the group of CCl4 injection. Our results demonstrate that 0.5 g/kg BW of TLP supplementation has a markedly protective effect against CCl4-induced liver injury; however, the protective mechanism and protective factors in the TLP remain to be explored.
