Effects of Vibrio alginolyticus on clotting factors and hemocyanin of white shrimp Litopenaeus vannamei / 溶藻弧菌影響白蝦凝血因子及血藍素之研究

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 94 / This study investigated the effects of bacterial cells and extracellular products (ECP) of Vibrio alginolyticus on clotting factors and hemocyanin of white shrimp Litopenaeus vannamei.
The oxyhemocyanin in hemolymph of white shrimp differed at different molt stage (A、B、C、D stage) with lowest concentration at A stage and highest at D stage. The changes of genetic expression (mRNA) of the coagulogen, transglutaminase and hemocyanin in hepatopancreas of white shrimp differed at different molt stage were investigated by using Real-Time PCR. The highest and lowest transglutaminase expression was found at D stage and B stage, respectively. The highest hemocyanin expression was found at D stage. Nevertheless, the coagulogen expression exhibited no significant difference at different molt stage.
The plasma of white shrimp was affected by pre-incubation or pre-chellenge with V. alginolyticus bacterial cells and ECP as visualized on the CIE gels. The coagulogen content was reduced following in vitro ECP treatment. In addition, the coagulogen content was reduced following in vivo bacterial cells and ECP treatment.
The changes of genetic expression (mRNA) of the coagulogen, transglutaminase and hemocyanin in hepatopancreas of white shrimp affected by bacterial cells and ECP of V. alginolyticus were investigated by using Real-Time PCR. The results revealed that mRNA expression of both coagulogen and transglutaminase was enchanced while the mRNA expression of hemocyanin was reduced post bacterial or ECP challenge.
The results of present study revealed that coagulogen was broken by V. alginolyticus causing the increase of mRNA expression of both coagulogen and transglutaminase in the shrimp, whereas the mRNA expression of hemocyanin was reduced that may be due to the destruction of hemocyanin synthetic pathway during the infection of V. alginolyticus.
