Two-stages culture of Haematococcus pluvialis and its astaxanthin components study / 雨生紅球藻兩階段養殖及其紅色類胡蘿蔔素(astaxanthin)組成之研究

The red carotenoid (astaxanthin) of microalga, Haematococcus pluvialis, was as feed additive for fish colorant of aquaculture, also has high antioxidant activation which can eliminate the free radical and stimulate immune responses for anticancer, therefore, it has high medical value. In the preliminary culture, it is found that JM is the optimal medium among the four medium (JM, OHM, Hydroponic, and Hyponex). In the first-stage culture shown that the maximum algal biomass reached on the 28th day (5.155×105 cells/mL) of culture, the contents of astaxanthin increases with the culture days increase, and the red color of algal body more and more dense as well. The unit of astaxanthin concentration reached to the maximum (2.7698×10-2 mg/mL) on the 210th day of culture. Using TLC and HPLC to analyze the astaxanthin of H. pluvialis and found it was composed by free, monoester and diester form of astaxanthin, in which monoester form was the most amount. In the second-stage culture, for stimulating the algal cell to alternate to the haematocyst, it is found that under 350 μE m-2s-1 of light density, the speedy of algal cells to turn red were slower in 3%NaCl than in 2%NaCl. But, in other light densities (150 μE m-2s-1 and 250 μE m-2s-1), the rapidest color alternation were all in 3%NaCl. However, its unit astaxanthin concentration is lower than those of 1%NaCl. Under 150 μE m-2s-1 light density with 1%NaCl resulted both the highest of unit astaxanthin concentration and its total content. Therefore, firstly increased the maximal algal biomass using the method of first-stage culture, and then in the second-stage culture, induced the algal cells turning red under optimal light densities and suitable concentration of NaCl, was suggested. This two-stages of H. pluvialis culture could be more economic and facilitate to promptly harvest large amount of astaxanthin.
