Incidences and Characterization of Streptococcus iniae Isolated from Commercical Tilapia in Taiwan / 魚型鏈球菌於市售吳郭魚之污染調查與其特性之探討

The objectives of this study were to investigate the incidences of Streptococcus iniae in commercial tilapia, and to characterize the growth and antibiotic subsceptibility of Streptococcus iniae isolates. Fifty samples of tilapia bought from traditional market and supermarket were analyzed the incidence rate of Streptococcus iniae by using the traditional biochemical test, API-20 Strep and PCR with Lox-1/Lox2 as primers. The incidence percentages of Streptococcus iniae in tilapia from the traditional market and the supermarket were 48% and 8%, respectively. For tilapia smaples from the traditional market and supermarket the detection rates for the presence of Streptococcus iniae in fish fins, surface, gill and eye position were 22%, 15%, 14%, 12%, respectively. All tested strains, including 63 fish isolates, 1 human isolate and 1 standrad strain showed β-hemolytic reaction on trypticase soy agar with 5% sheep blood. All strains showed the typical biochemical reactions of catalase (-), pyrrolidonylarylamidase (+), esculin hydrolysis (+) and VP (-). A DNA fragment with 870 bp was observed for all tested strains by PCR with Lox-1/Lox2 primers. After incubation of Streptococcus iniae ATCC 29178 in BHIB containing 0.5%, 1%, 1.5%, 2%, 2.5% and 3% NaCl for 10, 19, 25, 34, 53 and 73 hr, respectively, the cells reach the stationary phase. All Streptococcus iniae strains grew at 25℃ and 35℃, but not at 0℃ and 42℃. Most strains grew in 0.5~3% NaCl, but not in 4% NaCl. All strains can not growth at pH 4.5 and pH 10.5. All 34 isolates were sensitive to β-lactams, macrolides, chloramphenicol and vancomycin but only 7 (21%) isolates were sensitive to sulfamethoxazole/ trimethoprim. RAPD do not have the discriminatory power to differentiate the tested streptococcus isolates by using five different primers of ERICI, ERICII, M13R, 1290 and RAPD1.
