Characterization and biological functions of pleurocidin in grouper (Epinephelus coioides ) / 石斑魚抗菌蛋白基因選殖及生物功能

Pleurocidin, an antimicrobial peptide, plays important roles in fish to protect themself from microbial infection. Using a plaque screening assay, we isolated cDNA and genomic DNA clones encoding a novel pleurocidin wsa isolated from grouper (Epinephelus coioides) cDNA library and genomic DNA library. The pleurocidin cDNA consists of 519 bp with an open reading frame of 201 bp. The deduced protein has 67 amino acid residues, a calculated molecular mass of 7663 Da and an isoelectric point of 8.02. Either cleavable signal peptide or a nuclear localization signal (NLS) can be identified from the deduced sequence.Partial sequence analysis of 2 genomic clones (10,731 and 2951 bp) from the grouper pleurocidin suggrofthat in composed of multiple clustered genes. The grouper pleurocidin. Three genes are encoded in these clusters, but The sequences of three putative translated antimicrobial peptides geues shant minor differeces.The structures. Analysis of potential promoters and transcription factor binding sites implied that the immune-related transcription factor may be involved in pleurocidin gene regulation Staphylococcus coagulaes negatir、P. aeruginosa 、 Streptococcus pyogenes.
The amino acid sequences and gene organization of AMP are remarkably conserved in teleostean fisheo teleosts. Minimal inhibitory concentration (MIC) values of synthe fic grouper pleurocidin containine 21 amino acids against the Gram-positive bacteria. Grouper pleurocidin was expressed in at high levels in skin,intestine and kidney cells by real time PCR analysis,but much lese in liver, heart and muscle.
