碩士 / 長庚大學 / 生化與生醫工程研究所 / 95 / In this study we examined the behavior of freshly isolated rat hepatocytes cultivated within alginate and galactose-modified alginate gel beads. The hydrogel scaffold can mimic the in vivo extracellular matrix and provide a three-dimensional environment for cell proliferation and differentiation. Alginate modified with galactosamine, which provides a galactose moiety for recognition by hepatocytes surface receptor, was found to drastically up-regulate the metabolic functions of hepatocytes. Hepatocytes encapsulated in alginate beads were cultured in a pack-bed reactor and medium re-circulating speed in reactor was found to influence cell aggregate size, albumin secretion rates, and urea production rates.
