Extraction and functional analysis of polysaccharides from Chlorella pyrenoidosa / 小球藻多醣體的萃取與弁鉞?翿敦Q

碩士 / 嘉南藥理科技大學 / 化妝品科技研究所 / 95 / Polysaccharides are natural materials. They are most acquired from bacteria, ganoderma and higher plants. There are many studies of antitumor, immunostimulation and anti-virus have been discovered in the extremely high value of polysaccharides.
This study is designed to investigate the extract and purification of polysaccharides from Chlorella pyrenoidosa. The DPPH radical scavenging activities of the intracellular and extracellular polysaccharides are 70% and 16% for 1.5mg/mL, respectively. Trolox equivalent antioxidant capacity of the intracellular polysaccharides(1.5mg/mL)is 0.6mM and extracellular polysaccharides(1.5mg/mL)is 0.3mM . The inhibitory effect of lipid peroxidation in intracellular and extracellular polysaccharides are 63% and 10% for 1.5mg/mL, respectively. The antibacterial activity of the polysaccharide is also studied, it shows that the minimum inhibition and minimum bactericidal concentrations by intracellular and extracellular polysaccharides are (1) 125, 155 and 125, 155mg/mL respectively for Escherichia coli, (2) 125, 85 and 125, 85 mg/mL respectively for Pseudomonas aeruginosa, but only intracellular polysaccharides can do that for 16 and 31mg/mL for Propionibacterium ances, respectively. To determine the effectiveness and moisturizing ability of intracellular and extracellular polysaccharides, the test people were plastered with them and to detect after 20 minutes. The water content of skin nearly maintains up to 35% and 32%. As for inhibition of 5-Lipoxygenase activity, the IC50 of intracellular and extracellular polysaccharides are 0.10 ad 14 mg/mL, respectively. As for inhibiting the activities of matrix metalloproteinases, the intracellular polysaccharides are able to suppress the activities of MMP-2 at the treated concentration of 3、7mg/mL after 72 hours, but it is more obvious to do that after 96 hours. Then, the extracellular polysaccharides do not have this function.
As a whole, it elucidates that the polysaccharides of Chlorella pyrenoidosa except that of functions on antitumor and immunostimulation, this study also demonstrates that these extracts are capable of antioxidant, antibacterial, moisturizing, anti-inflammation and antiaging effects.
