The Study of Effects of Glucose Variation in Cultured Trypanosoma oreochromicus n. sp. / 葡萄糖變化對於吳郭魚錐蟲影響之研究

碩士 / 國立嘉義大學 / 水產生物學系研究所 / 95 / This experiment the glucose content to influence Trypanosoma oreochromicus n. sp., in grown、 morphology、 gene expression. Glycosomal glyceraldehyde phosphate dehydrogenase (gGAPDH) gene were used for phylogeny analysis and reconstructed phylogenetic tree.
Compare trypanosome was cultured in 10mM medium and 0mM glucose medium, total ammonia the trypanosome were 4.63×106 cells/ml、4.16×106cells/ml,respectively. In gene expression test, contrast ACTIN gene mRNA expression, hexokinase (HXK) genes not affecting glucose concentration change; glucose concentration change caused gGAPDH mRNA expression variation.
Partial glycosomal glyceraldehyde phosphate dehydrogenase (gGAPDH) gene was used in phylogenetic analysis of the trypanosome. The results of the topology analysis, there were highly significant in the body length、distance of midpoint of nucleus to posterior end、distance of midpoint of nucleus to anterior end. In phylogenetic analysis: used PCR cloned partial gGAPDH gene sequence. Array gGAPDH gene sequence, got a phylogenetic tree. Freshwater-fish parasite trypanosome, fall in the same clade, belong monophyletic.
