Preparation of Thermal Stable Microcapsulesby Using Fish Skin Gelatin as Carrie / 以魚皮明膠為載體製備耐熱性微膠囊

碩士 / 國立宜蘭大學 / 食品科學系碩士班 / 95 / This study is to develop thermal stable microcapsules to reduce the high temperature damage during thermal processing. Therefore, hydroxylpropylmethylcellulose (HPMC) and cross-linking agents were applied to enhance the thermal stability of fish skin gelatin (FG) and used as the wall materials of microcapsules prepared by spraying and phase separation methods. When HPMC was added in FG and then maturated: the absorbance (A400) significantly increased, which indicated the combinative gel was formed; the gel strength and storage modulus (G') increased, which indicated the gelation ability of combinative gels was enhanced; the melting temperature (Tm) and enthalpy increased, which indicated the thermal stability of combinative gel was improved. The FG/HPMC combinative gel and wax were then used as hydrophilic and hydrophobic phase, respectively. The microcapsules were prepared by spray drying and spray chilling for O/W and W/O emulsion, respectively. Though the latter exhibited higher water content, but thermal stability is still low. Therefore, phase separation method was suggested to prepare the thermal stable microcapsules. The salt-out caused phase separation of emulsion obtained by treatment with 1% sodium metaphosphate, floccules was then solidified with 2% TG. The melting temperature of microcapsules wall increased to 137℃. The microcapsules prepared by forming static electricity interaction between FG and polysaccharides by pH adjustment and 2% TG solidification. The thermal stability of solidified FG, Tm =143℃, was higher than that of without solidification. Moreover, when chitosan was added to the floccules of FG/polysaccharides complex and was solidified with TPP, the Tm was observed at 154℃. The highest thermal stable, Tm=159℃, of FG/HPMC combinative gel was obtained in wall material of multi-layer microcapsules prepared with W/O/W multiple emulsions by salting-out method.
