Analysis of N7-methylguanine and N7-ethylguanine in fish DNA by isotope-dilution with on-line SPE LC-MS/MS method / 運用連線固相萃取搭配液相層析串聯質譜儀與同位素稀釋法分析魚體DNA中甲基化與乙基化鍵結物

碩士 / 國立臺南大學 / 環境生態研究所碩士班 / 95 / In this study we use on-line solid-phase extraction (on-line SPE) liquid chromatography–tandem mass spectrometry (LC-MS/MS) with isotope-dilution to quantitatively analyze the DNA adducts of N7-methylguanine (N7-MeG) and N7-ethylguanine (N7-EtG) in small fish liver. The results show that N-nitrosodimethylamine (NDMA) induced an apparent linear dose-response for N7-MeG levels in liver tissue and a sublinear dose-response for N7-EtG levels was observed with increasing concentration of N-nitrosodiethylamine (NDEA). When fish received combined treatments of NDMA and NDEA, both N7-MeG and N7-EtG were dose-dependently increased. Moreover, the combined treatment of both agents did not generate significantly more N7-MeG and N7-EtG than either treatment alone. At the time-course experiment, N7-ethylated guanines were removed less efficiently than N7-methylated guanines. In the future, this method could be used at ecological studies to estimate the relationship between DNA alkylation and biological response from exposure to environmental alkylating agents.
