Immunostimulatory effects of sodium alginate and carrageenan on the immune ability and pathogenic resistance against Vibrio alginolyticus in the grouper Epinephelus coioides and Epinephelus fuscoguttatus / 藻酸鈉和鹿角菜膠激活點帶石斑和棕點石斑免疫能力與抵抗Vibrioalginolyticus感染之研究

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 95 / The respiratory burst, phagocytic activity, alternative complement hemolytic activity (ACH50) and lysozyme activity of orange-spotted grouper Epinephelus coioides were examined when the fish were injected intraperitoneally with sodium alginate at 10, 20, 30 mg kg-1 and iota ι-carrageenan at 10, 20, 30 mg kg-1, respectively after 24, 72 and 120 h. Serum ACH50 increased directly with dose after 24 and 72 h for both sodium alginate and ι-carrageenan treatments. The fish that received sodium alginate at 20 mg kg-1 after 24 and 72 h, and the fish that received ι-carrageenan at 30 mg kg-1 after 72 and 120 h showed significantly increased respiratory burst and phagocytic activity, respectively. In another experiment, E. coioides which had been injected individually with sodium alginate and ι-carrageenan at 10, 20, 30 mg kg-1, were challenged with V. alginolyticus at 1.8 × 1010 colony-forming units (cfu) fish-1 and then placed in seawater of 34‰. The survival of fish that received sodium alginate at 20 mg kg-1, and the fish that received ι-carrageenan at 30 mg kg-1 was significantly higher than that of fish which received saline and the control fish after 48 h as well as at the termination of the experiment (120 h after the challenge). It was therefore concluded that E. coioides which received sodium alginate at 20 mg kg-1 or ι-carrageenan at 30 mg kg-1 increased the innate immune response and resistance from V. alginolyticus infection.
Brown-marbled grouper Epinephelus fuscoguttatus which had been fed diets containing sodium alginate and kappa κ-carrageenan at 5, 10 and 20 g kg-1, respectively after 0, 2, 4, 8 and 14 weeks were examined for survival, growth, innate cellular and humoral responses as compared to the fish that fed control non-supplemented diet. Survial was 100 % for the fish that fed all diets after 14 weeks, and no significant difference in growth was observed among seven diets. The fish that fed a diet containing sodium alginate at 10 g kg-1, and fed a diet containing κ-carrageenan at 5 g kg-1 over 2-8 weeks showed significantly increased total leukocyte count, respiratory burst, phagocytic activity and index. In another experiment, E. fuscoguttatus which had been fed control diet, and all diets containing sodium alginate and κ-carrageenan after 14 weeks were challenged with V. alginolyticus at 5.0 × 109 cfu fish-1 and then placed in seawater of 34‰. The survival of fish that fed a diet containing sodium alginate at 10 g kg-1 and fed a diet containing κ-carrageenan at 5 g kg-1 was significantly higher than that of fish which fed the control diet over 96~168 h. It was concluded that E. fuscoguttatus which fed a diet containing sodium alginate at 10 g kg-1 or less, or fed a diet containing κ-carrageenan at 5 g kg-1 enhanced the innate immunity, and increased the resistance from V. alginolyticus infection.
