Molecular cloning and characterization of tilapia (Oreochromis mossambicus) follistatin gene and application of activin βA vaccine in tilapia infertility / 莫三鼻克吳郭魚卵泡抑素基因分子選殖與特性分析及啟動素βA疫苗應用於魚類不孕之研究

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 95 / Follistatin is a cysteine rich polypeptide with high conservation, belongs to follistatin family. Follistatin suppresses follicle stimulating hormone (FSH) secretion through its potent ability to bind and bioneutralize activin. In this study, we have cloned and characterized of the full-length of follistatin gene cDNA from tilapia (Oreochromis mossambicus). Tilapia full-length follistatin cDNA was 1081bp in length and could be deduced into 966 amino acids. The partial genomic DNA structure of follistatin was closed and which composed of five exons and four introns. The result of real-time quantitative PCR analysis showed follistatin gene expressed in brain, gill, muscle and ovary, highest expression was found in brain. Little expression of follistatin was detected in different embryonic developmental stages, and approaching sexual maturation phase follistatin expresion increased significantly. During reproductive cycle, follistatin gene expressed higher in previtellogenic stage, and decreased in late vitellogenic stage. On the contrary, activin mRNA was higher in perinucleoar oocytes, and became lower after entering vitellogenic stage., Higher follistatin mRNA and FSHβ mRNA was found in low temperature treatment. Low dose e stradiol inject could increase activin、FSHβ and follistatin mRNA expression. However negative feedback effect was found in estradiol high dose treatment. Low dose 17α-methyl-testosterone raised follistatin mRNA expression but supressed activin and FSHβ mRNA level. Chorionic gonadotropin elevate activin and FSHβ mRNA level. Recombinat human follistatin suppressed FSHβ mRNA expression in vitro significantly. No effect on endogenous follistatin mRNA level. The neurotransmitters, such as norepinephrine, acetylcholine chloride, dopamine and serotonin can increase follistatin mRNA level, but caffeine and GABA decreased follistatin mRNA level. To reduce FSHβ expression, Inject activin peptide vaccine can inhibit the development of oocytes and spermatogenesis and caused various infertility levels in tilapia.
