Effects of extracellular products of Vibrio harveyi and immunostimulants on clotting factors and hemocyanin of white shrimp, Litopenaeus vannamei / 哈威弧菌之細胞外產物及免疫刺激物對白蝦凝血因子及血藍素影響之研究

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 95 / This thesis first investigated the effects of extracellular products and products protease(22 kDa)of shrimp common pathogen--Vibrio harveyi on clotting factors and hemocyanin of white shrimp, Litopenaeus vannamei. In addition, β-glucan, Gelidium amansii hot-water extracts, sodium alginate and Fucoidan were served as immunostimulants(2g/kg feed)to feed white shrimp. Then the changes of genetic expression of the coagulogen, transglutaminase and hemocyanin in hepatopancreas of white shrimp were studied.
In the in vivo study, the plasma, coagulogen and hemocyanin of white shrimp was affected by pre-challenge with V. harveyi ECP and purified protease as visualized on CIE gels, the coagulogen content was reduced and the hemocyanin was not affected following ECP and protease treatment. Using the Real-time PCR technique, the mRNA expression of both coagulogen and hemocyanin was reduced post ECP and protease challenge, but the transglutaminase reduction could only be caused by ECP.
On the immunostimulants study, white shrimp fed with Fucoidan, Gelidium amansii hot-water extracts, and β-glucan all exhibited an increase of coagulogen expression. Shrimp fed with Fucoidan and β-glucan also exhibited an increase of transglutaminase expression. In addition, shrimp fed with exhibited an increase of hemocyanin expression.
The results of present study reveal that the coagulogen, transglutaminase and relative factors of white shrimp, must exhibit clotting and immune reaction as challenge by ECP and protease, result in their content reduction, may affect their synthetic pathway leading to reduction of their expression. Hemocyanin content was not affected by ECP and protease. However, its synthetic pathway factors may be affected, since the mRNA expression of hemocyanin was reduced. Therefore, the use of immunostimulants may increase the non-specific immunity such as the increase of gene expression coagulogen, transglutaminase and hemocyanin and thus enhance the resistance against the infection of Vibrio harveyi.
