Anti-atherosclerosis Function and Machanism of Astaxanthin Using Cultured Cell model / 以細胞培養模式探討蝦青素抗粥狀動脈硬化之功能與機制

碩士 / 國立臺灣海洋大學 / 食品科學系 / 95 / Astaxanthin, a red-orange carotenoid pigment, is a powerful biological antioxidant and provides anti-inflammatory activities that occurs naturally in a wide variety of living organisms. Atherosclerosis is a chronic inflammation disease, it can influence human’s heath. Atherosclerosis can’t be defined as a single disease entity. Atherosclerosis possessed of the great relation with oxidative stress and inflammation had be major causes. In some early study, it shown that astaxanthin could inhibit the possession of atherosclerosis and improved plaque stability in vivo. However, detailed machanisms have not been fully delineated. In this research, we interested in evaluation that whether astaxanthin could regulate monocyte adhesion to endothelial cell and formation of foam cells. 100 □M astaxanthin could inhibit of TNF-α (tumor necrosis factor-α) stimulated human monocyte THP-1 adhesion to human umbilical vein endothelial cells (HUVEC) (from 4-fold to 1.4-fold) that mediated by cellular adhesion molecular VCAM-1 (from 47% to 38%) and E-selectin (from 68.6% to 39.1%), also inhibited TNF-α-induced monocyte chemoattractant protein (MCP-1) (from 3.8-fold to 1-fold) and interleukin-8 (IL-8) (from 1.8-fold to 1.3-fold) mRNA expression in HUVEC. 100 □M astaxantin influenced murine macrophage J774.1 ability to endocyte oxLDL that was formation of macrophage-derived foam cells through down-regulation of oxLDL receptor, SR-A (from 50.1% to 36.8%) and CD36 (from 23.9% to 19.1%), it might be due to scavenge production of intracellular ROS (from 130% to 108%). Therefore, this study suggested that astaxanthin could block monocyte adhesion to endothelium by reduced cell adhesion molecules and chemokines expression and inhibit formation of foam cells through down-regulation of oxLDL receptor expression in anti-atherosclerosis function and mechanism using cultured cell model.
