Acute toxicity in zebrafish larvae after exposure to flavonoids / 類黃酮對斑馬魚稚魚之急毒性

碩士 / 中臺科技大學 / 醫學生物科技研究所 / 96 / Biflavonoids and monoflavonoids, naturally occurring flavonoids, widely present in many plants. In recent years, many studies indicated that biflavonoids and monoflavonoids have various pharmacological and physiological effects. In order to identify the ability of these biflavonoids and monoflavonoids to inhibit cell proliferation, we developed that zebrafish larvae as a model system for the developmental toxicity study of the flavonoids and their chemical derivatives. Both chemical and genetic approaches to pathway analysis are increasing our understanding of flavonoids function and how it relates to human disease. Thus, it might be advantageous to test drugs and its function in model organisms to predict the corresponding effects in humans.

Zebrafish larvae were exposed to different concentration of biflavonoids including Amentoflavone and Hinokiflavone and monoflavonoids including Flavanone, 6-Fluoro-3-methoxyflavanone and 3,6-Difluoroflavanone. After demonstrated developmental toxicity with LC50, we then use TUNEL assay to test whether apoptosis or not and further detection caspase-3,caspase-9 and Hsp90 activity. TUNEL assay indicated zebrafish larvae after exposure to flavonoids have different levels of apoptosis. Caspase-3, caspase-9 activity assay and and Hsp 90 relative abundance indicated zebrafish larvae elevated caspase-3, caspase-9 and Hsp90 activity after exposure to flavonoids. These data clearly demonstrate a role for acute toxicity in zebrafish larvae after exposure to flavonoids. Nevertheless, the detailed mechanisms must to be elucidated by more experiments, ex. microarray and 2D electrophoresis analysis.
