Cloning and Application of Zebrafish MSTN and GDF-11 Promoters / 斑馬魚MSTN及GDF-11啟動子之選殖與應用

碩士 / 國立彰化師範大學 / 生物學系 / 96 / Myostatin (MSTN), or Growth and Differentiation Factor-8 (GDF-8) is a member of the transforming growth factor-β superfamily( TGF-β superfamily ) that negatively regulates growth of skeletal muscle tissue. The mechanisms by which this molecule mediates its effect in vivo are poorly understood. Whereas myostatin (MSTN) in mammals is almost exclusively expressed in the muscle by a single encoding gene, zebrafish possess two MSTN genes which are differentially expressed in both muscular and non-muscular tissues. GDF11 is hypothesized to be a key regulator of axial skeleton patterning in developing vertebrates. Besides, it was also demonstrated to inhibit olfactory epithelial neurogenesis. This negative autoregulatory action of GDF11 is strikingly like that of its homolog, GDF8/myostatin, in skeletal muscle, suggesting that similar strategies establish and maintain proper cell number during neural and muscular development. Using zebrafish as an animal model for drug screening, the effects of TSA, VPA, curcumin and crude extracts of Morus alba L on MSTN-1, MSTN-2 and GDF-11 gene expression were evaluated by reverse-trascription polymerase chain reaction (RT-PCR) and promoter activities assy. The promoter region of the zebrafish MSTN-1, MSTN-2 and GDF-11 was cloned from a genomic DNA library and sequenced, and its activity was further characterized using green fluorescent protein (GFP) transient transfection expression system. The results showed that both find zebrasfish MSTN-1, MSTN-2 and GDF-11 promoter region displayed weak promoter activity, curcumin up-regulated MSTN-1, MSTN-2 and GDF-11 gene expression and VPA possible to dowm-regulated MSTN-2 gene expression. These promising data open the way to new therapeutic approaches in muscle or neuron diseases through targeting of the myostatin pathway.
