碩士 / 國立彰化師範大學 / 生物學系 / 96 / At the nucleus of eukaryocytes, DNA winds around histones to form the neucleosome. Histone acetyltransferase, HAT, will acetylate histones and then the acetylated histones will leave DNA due to gene expression. On the contrary, histone deacetylase, HDACs, will remove acetyl group to inhibit gene expression. Trichostatin A is a HDACs inhibitor which can stop the cell cycle to induce apoptosis. In addition, another HDACs inhibitor, Nicotinamide, could increase gene expression to promote aging of yeast. This research is going to evaluate the effects of zebrafish embryo development on Trichostatin A and Nicotinamide and to screen natural materials which can affect zebrafish development. The results showed: Trichostatin A caused neck deformation of the embryo, but not obvious after 4 hrs of fertilization. However, Nicotinamide influenced spinal development of embrylo in different developmental stages. The embryo deformation induced by inhibition of HDACS class I, II and III simultaneously was much more serious than that of Trichostatin A or Nicotinamide. The data also showed that A025 extract caused the failure of yolk absoption and deformation of zebrafish embryo. At last, this research also established the fluorescent screening system of HDACs inhibitor to perform microinjection.
