碩士 / 國立東華大學 / 海洋生物科技研究所 / 96 / In continuing search the bioactive secondary metabolites from marine microorganism, we found that the extracts of marine cyanobacteria Lyngbya majuscula, collected from Ryukyu Island, showed significant anticancer and anti-dialetes activities. Through bioassay-guided separation, we isolated four malyngamides, including three new compounds, malyngamide Z (1), malyngamide X (2), malyngamide 1 (4) and a known compound, isomalyngamide A (3). We also isolated four aplysiatoxins, including one new compound, 3-methoxy debromoaplysiatoxin (7) and three known compounds, ahhydrodebromoaplysiatoxin (6), debromoaplysiatoxin and oscillatoxin B (8) from L. majuscula. The structure of these compounds were determined by 1D and 2D NMR spectroscopic analysis.
In the biological test, compound 1、3 and 5-8 exhibited activity in cell lines of human hepatoma 2 (Hep2) and human colorectal cancer (WiDr). The ED50 values were between 11.6 ~ 26.0µg/ml. In brine shrimp toxicity test, malyngamides (1、3、4) showed no activity and aplysiatoxins 5 ~ 8 showed strong activity. The LD50 values were between 0.5 ~ 7.7 µg/ml.
