碩士 / 國立高雄海洋科技大學 / 水產食品科學研究所 / 96 / The lectins and polysaccharides wildly exist in legumes plants, edible mushrooms, fungus and marine alga. The potential biological activity of lectins and polysaccharides from marine alga, the cheap resources, will provide an excellent food for promotion of human health. In present study, the isolated crude lectins and polysaccharides from Sargassum duplication and the purified lectins from Codium fragile were used to stimulate the human peripheral blood monocytes cells (PBMNC) against the growth of leukiemia.
Our results have indicated that five fractions from cold water extract of Sargassum duplication were obtained by different degree of saturated ammonia sulfate. The AS-2 and AS-3 fractions exerted the greatest hemagglutining activity, up to 22-23 titers. The AS-4 fraction, with the lowest hemagglutining activity, exerted the greatest immunomodulatory effects against U937 leukemia cell line. The cold water crude extract was separated by Amicon (cut off at 10 kDa) to obtain the most active fraction as less than10 kDa. This fraction exist the greatest immunomodulatory effects against U937 leukemia cell line up to 36.4% at 100 μg/mL. Further separating procedure, DEAE sepharose ion-exchange chromotagraphy, performed to get two immunomodulatory fractions, F1-280 and F2-490. the immunomodulatory effects of F1-280, full of carbohydrate, and F2-490, full of protein, against U937 leukemia cell line were about 64.4% and 68.5%, respectively. The purified lectin from Codium fragile exist immunomodulatory effects against U937 leukemia cell lines, however, also exist strong hemagglutining activity, up to 22-23 titers. Collectively, the lectins and polysaccharides existed in marine alga exhibit immunomodulatory effects
against U937 leukemia cell lines.
