碩士 / 國立屏東科技大學 / 水產養殖系所 / 96 / The object of this study was to detect temporal genetic variations of Japanese eel, Anguilla japonica. Temporal genetic fluctuation of Japanese eel elvers in the Tanshui Estuary, northern Taiwan from Dec. 1999 to Feb. 2000 and from 1989 to 2008 were examined using mitochondrial DNA and microsatellite loci. Mean pairwise nucleotide diversity of mtDNA between nine yearly sample ranged from 0.008±0.005 (2000, 2008) to 0.019±0.006 (2005). Mean pairwise nucleotide diversity between sample from Dec. 1999 to Feb. 2000 ranged from 0.008±0.004 (Jan. 2000) to 0.015±0.003 (Feb. 2000). This result did not conform to genetic isolation by time. The fluctuations of nucleotide diversity within nine yearly samples expressed a significantly similar pattern with the annual catch of elvers. The means observed heterozygosity (Ho) within yearly samples from eight polymorphic microsatellite loci ranged from 0.419±0.225 (2003) to 0.617±0.159 (2005). The Ho fluctuations of yearly samples have a significantly similar pattern with the annual catch of elvers during 1989 and 2008. The significant differentiation was determined from all yearly and monthly samples (Fst=0.027, Fst=0.032, p<0.01). The genetic diversity of Japanese eel samples occurred yearly fluctuation which was not extended by prolong temporal distance. To evaluate effective population size (Ne) of Japanese eel using Markov chain Monte Carlo (MCMC) method. Ne was decline from 21379 (1989) to 8709 (2008). The population genetic structure of Japanese eel is an important indicator to assay fishery resources. For sustainable management of Japanese eel fishery, we should continuously detected the genetic fluctuation in the future.
