Culture of green flagellate, Euglena geniculata Duj / 膝曲裸藻之培養條件探討

碩士 / 國立屏東科技大學 / 水產養殖系所 / 96 / Euglena geniculata Duj is a kind of protozoans which has been investigated for its potential use as live food for the larvae of Giant freshwater prawn, Macrobrachium rosenbergii. In the present study, the effects of salinity and pH on the tolerance of E. geniculata Duj was investigated as a fundamiental study to determine the optimum culture conditions. The results showed that the salinities tolerance ranged between 0-15 psu, pH tolerance ranged between 3-8.
In the first experiment, we investigated three levels of temperatures (25, 30 and 35 ℃), four levels of salinities (0, 10, 15 and 30 psu) without or with carbon dioxide (air or air + CO2) addition effect on growth and survival of E. geniculata, under laboratory conditions. The results showed that the highest production was 35 ℃, followed by 30 ℃ and the lowest was 25℃.The optimum salinity conditions were 0 psu for growth and no survival at 30 psu. The growth peak was on the sixth day without carbon dioxide and droped, otherwise on the eleven day with carbon dioxide and then decreased.
The growth and survival of E. geniculata in comparison to other species used for feeding the larvae of the Giant freshwater prawn, M. rosenbergii were preformed. In the second experiment, the larvae were fed with protozoan (E. gracilis), four kind of microalga (Isochrysis galbana tml, Chaetoceros muelleri, Tetraselmis chui and Nannochloropsis oculata), and Artemia salina from the stage of zoea 1 (Z 1) to zoea 11 (Z 11) were conducted. The survival of development, body length of zoea (Z 11) of M. rosenbergii fed with the E. geniculata are compared to those obtained with the microalga and Artemia salima. The mortality were 100% among microalga in the seventh day. The similar good effects were on the survival and growth of the larvae of M. rosenbergii between E. geniculata and Artemia salina, however, the quantity of ammonia-nitrogen (TAN) and nitrite-mitrogen (NO2--N) had a significantly difference (p<0.05).
