Molecular cloning of alpha2-macroglobulin from the scalloped spiny lobster Panulirus homarus and its expression following Vibrio alginolyticus injection / 波紋龍蝦α2巨球蛋白分子選殖及注射溶藻弧菌的表現

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 96 / Alpha2-macroglobulin (α2-M) is a high molecule, and abundant in vertebrates and members of several invertebrate phyla. α2-M neutralizes pathogenic proteases that contributes to pathogen virulence.
The objective of this study is aimed to clone α2-M cDNA sequence from the haemocytes of scalloped spiny lobster Panulirus homarus and its tissue expression level. Another target is to measure the expression level when lobster is injected with Vibrio alginolyticus.
P. homarus α2-M cDNA consists of 4809 bp with 72-bp 5`UTR, 357-bp 3`UTR and 4380-bp open reading frame (ORF) which encode 1460 amino acid. The predicted molecular mass of the translated protein is 161.2 kDa, with an estimated pI of 5.73. The α2-M deduced amino acid of P. homarus shows similarity of 68% to that of Macrobrachium rosenbergii. P. homarus α2-M is strongly expressed in haemocytes but not in other tissues.
The expression level of P. homarus α2-M increased after 6 h following V. alginolyticus injection indicating activation by V. alginolyticus.
