Assessment of collagen extracts from swim bladder on osteoblast and anti-inflammation using cultured cell model / 以細胞培養模式評估魚鰾膠原蛋白萃取物對於造骨細胞以及抗發炎之影響

碩士 / 國立臺灣海洋大學 / 食品科學系 / 96 / Osteoporosis is a degenerative bone disease of bone that leads to an increased risk of fracture, and characterized by a reduction in bone mineral density due to a loss of calcium and collagen. Recent studies indicate that collagen possessing a wide range of biological activities including curing rheumatoid arthritis and prevent osteoporosis. Collagen was usually found in chicken and bovine. However, there are misgivings, for example: Avian Influenza and Bovine Spongiform Encephalopathy. Recent research reaved that inflammatory and osteocytes play an important role in osteoporosis. In the first part of this study, collagen was extracted form swim bladder by NaOH, HCl and pepsin. The collagen content was 21%. Second, by using inflammatory inhibitory model of RAW264.7 cell, collagen showed the inhibiton of NO production of RAW264.7 cocultured by lipopolysaccharide (LPS), and the decrease of iNOS and COX-2 gene expression, Further use RT-PCR reveal that collagen inhibit the expression of proinflammatory cytokine (IL-1��, IL-8). The effect of scavenge ROS we also found by using FACS assay and fluorescence stereomicroscope. In osteocytes study, we found that collagen stimulated an increase in alkaline phosphatase (ALP) activity and mineralization on MG-63, a human osteoblast cell line.
