Isolation of bacteria and effect of sulfur metabolism in the hydrothermal vents surrounding Kueishan Island. / 龜山島熱泉週邊海域細菌之分離及其硫代謝能力分析

碩士 / 海洋大學 / 環境生物與漁業科學學系 / 96 / The average temperature, salinity, pH and sulfide were 25.1℃, 33.78 psu, pH 7.9 and 0.8 μM measured from surrounding of the Kueishan island in 2005. However, the temperature, pH and sulfide were 29.4℃, pH 5.3 and 667.52 μM in the hydrothermal vents. The bacteria took an important roles in this special environment.
This kind of bacteria was cultivated in middle temperature condition. Isolate strains were selected to study their morphologies and physiological characteristics. These similar bacteria were compared by using the rank of 16S rRNA. The D-1 similar bacteria was Dietzim, 9-2 similar bacteria was Marinobacter, 2-11 was Rhodococcus and 12-4 was Salinicoccu. Then the isolate strains was observed the sulfur metabolism.
The most notable reaction in each bacterium of sulfide concentration was Rhodococcus sp. It produced the highest sulfide concentration in 36 hrs. The others produced the lower sulfide concentration.
Rhodococcus sp., Dietzia sp., Marinobacter sp. and 9-3 sp all could oxidize the sulfur and produce sulfate. The metabolism concentration of sulfate during 96 hr were 84.2, 63.8, 100.3 and 132.6 μM.
This study also discovered the Rhodococcus sp. had the disproportionation function in 6-36 hr. This results indicate the Rhodococcus sp. could reduced the sulfur to sulfide and oxidize the sulfur to sulfate at the same time.
